Specificity

Clone 1H5 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects PAK-1. It targets an epitope within 15 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the N-terminal half of human PAK-1.

Application

Quality Control Testing

Evaluated by Western Blotting in C6 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PAK-1 in C6 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PAK-1 in Human brain and Rat brain tissue lysates.

Affinity Binding Assay: A representative lot of this antibody bound PAK-1 peptide with a KD of 1.3 x 10-6 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PAK-1 in human stomach tissue sections.

Target description

Serine/threonine-protein kinase PAK 1 (UniProt: EC:2.7.11.1, Alpha-PAK, p21-activated kinase 1, PAK-1, p65-PAK) is encoded by the PAK1 gene (Gene ID: 5058) in human. PAK-1 is a serine/threonine protein kinase that is involved in intracellular signaling pathways downstream of integrins and receptor-type kinases. It is in active conformation in its monomeric form and is autoinhibited upon homodimerization. It plays an important role in cytoskeleton dynamics, cell adhesion, migration, proliferation, and apoptosis. It is also involved in vesicle-mediated transport processes. It is involved in the reorganization of the actin cytoskeleton, actin stress fibers and of focal adhesion complexes. Its protein kinase domain is localized in amino acids 270-521. PAK-1 is activated by interaction with Cdc42 and Rac1. It functions as a GTP effector that links Cdc42 and Rac1 to JNK MAP kinase pathway. It phosphorylates and activates MAP2K1, and thereby mediates activation of downstream MAP kinases. It is reported to directly phosphorylate BAD and protect cells against apoptosis. In cells undergoing apoptosis, its activity is inhibited due to binding of CDK11A and CDK11B. PAK-1 is reported to phosphorylate RAF1 at serine 338 and 339 that results in RAF1 activation and translocation to mitochondria where it can phosphorylate BAD. PAK-1 is activated following autophosphorylation at threonine 423 in response to a conformation change that is triggered by interaction with GTP-bound Cdc42 or Rac1. Binding of GTP-boundCdc42 or Rac1 to its autoregulatory region (aa 70-140) releases monomers from the autoinhibited homodimeric state and enables phosphorylation (at Thr 423) and activation. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
