General description

ENPEP gene encodes the protein aminopeptidase A (APA), which is a membrane-bound ectopeptidase belonging to the zinc metalloprotease family. APA is expressed in a wide range of tissues, including the brain. Human ENPEP is localised to the chromosomal region 4q25. Human APA expressed in kidney is called gp160 and has a molecular weight of 160kDa.

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. Biochem/physiol Actions

APA (aminopeptidase A) that is encoded by ENPEP gene cleaves acidic residues, such as aspartate and glutamate, from the N- terminal of its substrates and its activity is modulated by calcium levels. It is one of the key enzymes in the central renin-angiotensin system, and regulates hypertension. It converts angiotensin II to angiotensin III by cleaving the N-terminal aspartate residue. gp160 plays a key role in the development of kidney, and therefore is also known as the human kidney differentiation antigen. APA activity has been shown to be decreased in various renal cell carcinomas as compared to normal renal cells. This might lead to an increase in the level of angiotensin II in renal carcinomas, hence contributing to angiogenesis. It has been suggested that the formation of Amyloid β peptides is facilitated by APA, where it truncates the full length Amyloid β at the N-terminal.
