General description

El medio de gradiente de densidad OptiPrep, también conocido como yodixanol, es un medio de gradiente de densidad yodado no iónico de baja osmolaridad. Es una disolución estéril y comprobada para endotoxinas que contiene yodixanol al 60 % (p/v) en agua y está diseñada para el aislamiento in vitro de partículas biológicas. Se puede utilizar para el fraccionamiento y la separación de orgánulos celulares y subcelulares.[1]

Application

OptiPrep se ha utilizado para aislar moléculas, células y orgánulos subcelulares, como ADN, citocinas, lisosomas, retículo endoplásmico, peroxisomas, linfocitos T y linfocitos citolíticos naturales.[1] También se ha utilizado para las siguientes aplicaciones:

  • investigar la eficiencia de la encapsulación unicelular.[1]
  • el aislamiento basado en gradiente de las células cancerosas senescentes.[2]
  • el aislamiento de viriones no encapsulados de los lavados broncoalveolares.[3]
  • el aislamiento de lipoproteínas y macromoléculas
  • purificación de viriones
  • purificación de células neuronales

  6. Why choose Product D1556, OptiPrep, instead of other density gradients such as Percoll or Histopaque?

    1. Percoll and Histopaque must be removed prior to use through washing.  If Percoll and Histopaque are not removed, these density gradients can be toxic to mammalian cells.  In comparison, OptiPrep has no potential for bioaccumulation and can be considered nontoxic to cells. OptiPrep also has lower viscosity as compared to polysucrose based products such as Histopaque or Ficoll.

    Expensive OptiPrep

    It worked as OptiPrep from other companies, but it's over-priced, in my opinion.
