Gene Information

human ... NANOG(79923)

General description

Nanog is a transcription factor homeoprotein that is involved in maintaining embryonic stem cell pluripotency and self-renewal. Embryonic stem cells with a deleted Nanog gene lose pluripotency. Conversely, embryonic stem cells that are forced to differentiate will downregulate Nanog expression. Nanog expression has been shown to be absent in normal adult organ tissues, but positive expression in undifferentiated germ cell tumors such as seminoma, dysgerminoma and embryonal carcinoma has been noted. Therefore, Nanog may be used as an aid in the determination of undifferentiated tumors of germ cell origin from non-germ cell tumors.

The embryonic stem cell transcription factors SOX2, NANOG, and OCT3/4 are involved in the regulation of germ cell tumor growth and differentiation. They, and D2-40 (podoplanin), an antigen expressed in seminomas, are emerging as useful diagnostic markers in testicular germ

The NANOG gene, a member of the homeobox family of DNA binding transcription factors, was recently identified in a screen for pluripotency-promoting genes. NANOG overexpression in murine embryonic stem cells is sufficient to maintain self-renewal and to block differentiation. The

Embryonic stem (ES) cells undergo extended proliferation while remaining poised for multilineage differentiation. A unique network of transcription factors may characterize self-renewal and simultaneously suppress differentiation. We applied expression cloning in mouse ES cells to isolate a self-renewal determinant. Nanog

Several extrinsic signals such as LIF, BMP and Wnt can support the self-renewal and pluripotency of embryonic stem (ES) cells through regulating the "pluripotent genes." A unique homeobox transcription factor, Nanog, is one of the key downstream effectors of these

Embryonic stem (ES) cells derived from the inner cell mass (ICM) of blastocysts grow infinitely while maintaining pluripotency. Leukemia inhibitory factor (LIF) can maintain self-renewal of mouse ES cells through activation of Stat3. However, LIF/Stat3 is dispensable for maintenance of
