General description

Beta-arrestin-2 (UniProt P32121; also known as Arrestin beta-2) is encoded by the ARRB2 (also known as ARB2, ARR2, BARR2) gene (Gene ID 409) in human. β-Arrestin 1 and 2 are cytosolic adapter proteins originally identified as negative regulators of G protein-coupled receptor (GPCR) signaling through receptor desensitization and internalization. It is now known that these multifunctional scaffold proteins also elicit G-protein-independent signalling pathways by linking the activated GPCRs to multiple signaling molecules, such as ERK1/2, Akt, and Src kinases. In addtion to GPCRs, β-arrestins are also involved in receptor tyrosine kinase (RTK) signalling pathways, and β-arrestin-2 is reported to link Src and Akt to insulin receptor (IR) to ensure a full Akt activation. Although β-arrestin-1 and β-arrestin-2 share a high degree of structural homology, they do display specific functions in several physiological processes. β-arrestins can adopt multiple conformations to display distinct sets of interacting domains. Recruitment of β-arrestins to a specific GPCR results in the formation of distinct β-arrestin scaffolds, allowing a high level of specificity for a given agonist.

Specificity

Expected to react with spliced isoforms 1, 4, and 5 reported by UniProt (P32121). Reactivity toward spliced isoforms 2 & 3 has not been determined.

Immunogen

Epitope: C-terminal region.
KLH-conjugated linear peptide corresponding to a sequence in the C-terminal region of human beta-Arrestin-2.

Application

Anti-beta-Arrestin-2 Antibody, clone 5E12.1 is an antibody against beta-Arrestin-2 for use in Western Blotting, Immunohistochemistry (Paraffin).
Immunohistochemistry Analysis: A 1:50-250 dilution from a representative lot detected beta-Arrestin-2 in rat testis and human cerebral cortex & liver tissues.

Quality

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected beta-Arrestin-2 in 200 µg of HeLa cell lysate.

Target description

~52 kDa observed. 46.11 kDa (isoform 1), 47.27 kDa (isoform 2), 45.56 kDa (isoform 3), 48.02 kDa (isoform 4), and 4439 kDa (isoform 5) calculated.
