Specificity

Clone 1G10 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects IL-12A. It targets an epitope within 18 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 18 amino acids from the N-terminal half of human IL-12A.

Application

Quality Control Testing

Evaluated by Western Blotting in THP-1 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected IL-12A in THP-1 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000-10,000 dilution from a representative lot detected IL-12A in lysate from human liver tissue and Raw 264.7 cells .

Flow Cytometry Analysis: 0.1 µg from a representative lot detected IL-12A in one million THP-1 cells.

Affinity Binding Assay: A representative lot of this antibody bound IL-12A peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Interleukin-12 subunit alpha (UniProt: P29459; also known as IL-12A, Cytotoxic lymphocyte maturation factor 35 kDa subunit (CLMF p35), IL-12 subunit p35, NK cell stimulatory factor chain 1 (NKSF1)) is encoded by the IL12A (also known as NKSF1) gene (Gene ID: 3592) in human. IL-12A is a cytokine that, along with IL12B, forms a disulfide-linked heterodimer interleukin IL-12. IL-12 alpha functions as a regulator of T cell mediated cytotoxicity, lymphocyte proliferation and activation of natural killer cells enhancing their cytotoxic activity. IL-12A is primarily localized in the cytoplasm of producing cells and is secreted into the extracellular space as part of the IL-12 heterodimer. The secretion process involves the Golgi apparatus for processing and packaging before release. IL-12A is predominantly expressed by antigen-presenting cells (APCs) such as dendritic cells, macrophages, and B cells. Its expression can be induced by various stimuli, including microbial products, antigenic stimulation, and the presence of other cytokines (e.g., IFN- ). All forms of IL-12 bind to the IL-12R which includes IL-12RbI and IL-12RbII subunits expressed on T cells and natural cells. IL-12 stimulates the production of IFN- and TNF- , subsequent production of nitric oxide that leads to pathogen destruction. Reports suggest the involvement of JAK/STAT pathway components in mediating these effects. The effects of IL-12 have been implicated in bacterial, fungal and viral infections. IL-12 is critical for controlling infections by intracellular pathogens (e.g., Mycobacterium tuberculosis). Studies have indicated that IL-35 plays an important role in controlling immune-related disorders, including autoimmune diseases, infectious diseases, and cancer. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
