General description

Cathepsin S (CTSS) is a lysosomal cysteine endopeptidase of the papain family or C1 family. It is produced as an inactive preproenzyme which is then converted into the active form in lysosomes and late endosomes.

Biochem/physiol Actions

Cathepsin S (CTSS) is a key participant in MHC (major histocompatibility complex) class II-mediated immune responses, where CTSS degrades Ii (type II membrane glycoprotein invariant chain) sequence of MHC II aβ heterodimers. Ii contains signaling motifs and its degradation leads to successful association of class II associated invariant chain peptide (CLIP) with the peptide groove of the MHC class II molecule. This is essential for antigen loading on MHC II complex during antigen presentation. Overexpression of this protein in mice leads to increased proteinase-activated receptor-2 (PAR-2) expression in dendritic cells resulting in scratching behavior and Th1 (T-helper) cell-associated cytokine expression. This can eventually lead to chronic atopic dermatitis in mice.
