General description
NEK11 is a member of the NEK family and the protein localizes to the nucleoli where it may function with NEK2A in the S-phase checkpoint. NEK2 phosphorylates NEK11 in its C-terminal autoinhibitory domain and elevates NEK11 kinase activity by dissociating the autoinhibitory domain from the N-terminal catalytic domain. NEK11 protein appears to play a role in DNA replication and response to genotoxic stress. NEK11 activity is activated in HeLa cells treated with various DNA-damaging agents and replication inhibitors. Activation of NEK11 by genotoxic agents is suppressed by inhibitor of ATR/ATM-mediated signalling indicating that NEK11 has a role in S-phase checkpoint downstream of ATR/ATM signalling.
