Biochem/physiol Actions

Aib-1 is an inhibitor of β-Amyloid oligomerization. Aib-1 is an endomorphin analog more stable to enzymatic degradation and incorporating the β-breaker 2 aminoisobutyric acid. It interacted with Aβ and markedly inhibited the formation of toxic oligomer and fibril growth using in vitro assays, prevented Aβ toxicity in neuronal PC12 cells, and increased longevity in a fly model of Alzheimer′s disease. Aib-1 activity is thought to be due to multi-mode interactions including aromatic, hydrophobic, and polar contacts with Aβ.
