General description

Histone H3 is one of the five main histone proteins that are involved in the structure of chromatin in eukaryotic cells. Histone H3 features a main globular domain and a long N-terminal tail. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Histone H3 is specifically phosphorylated during both mitosis and meiosis in patterns that are specifically coordinated in space and time. These phosphorylations are initiated at different phases of the cell division. Histone H3 variants (H3.1, H3.2 and H3.3) have been implicated in the epigenetic memory of cellular state. Genome-wide patterns of H3 are dependent on amino acid sequence and change with cellular differentiation at developmentally regulated loci.

Specificity

Expected to react with most other species based on extreme conserved homology among species.

Immunogen

Epitope: Trimethylated (Lys4)
Synthetic peptide corresponding to residues surrounding and including trimethlyated Lys 4 of Histone H3.

Application

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected trimethyl-Histone H3 (Lys4) in A431 and NIH/3T3 cells.
The unconjugated version of this antibody has been shown to work in ChIP, DB, WB, ChIP-seq.
