Specificity

Clone 1C14 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects human Mn-SOD.

Immunogen

His-tagged full-length mature (without the transit peptide) recombinant human Mn-SOD.

Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Mn-SOD in HepG2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Mn-SOD in HeLa cell lysate.

Affinity Binding Assay:: A representative lot of this antibody bound Mn-SOD proteins with a KD of 4.5 x 10-8 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Mn-SOD in human colon tissue sections.

Anti-Mn-SOD, clone 1C14 ZooMAb®, Cat. No. ZRB1246, is a recombinant Rabbit monoclonal antibody that specifically targets Mn-SOD and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Superoxide dismutase [Mn], mitochondrial (UniProt: P04179; also known as EC:1.15.1.1, Mn-SOD) is encoded by the SOD2 gene (Gene ID: 6648) in human. Superoxide dismutases (SODs) provide defense against reactive oxygen species. Three types of SODs are found in organisms and are named based on the nature of the metal ion(s) found at their active sites. The Cu/Zn superoxide dismutase (Cu/ZnSOD), is primarily localized in the cytosol, nucleus, peroxisome, and intermembrane of mitochondria in eukaryotes. The Mn-SOD is found in the mitochondria, and the Fe-SOD is found primarily in bacteria. Mn-SOD is a homotetrameric enzyme, localized in the mitochondrial matrix, catalyzes the dismutation of superoxide radicals in mitochondria by converting anion superoxide into hydrogen peroxide and oxygen. Each monomer of Mn-SOD contains an active site bound to manganese (Mn) ion. Mn-SOD constitutes about 10 15% of the total SODs. The Mn ions can change between its 3+ and 2+ oxidation states within the dismutase mechanism that consists of two half-reactions. Electron exchange in metal complexes is reported to occur by either an inner-sphere or outer-sphere mechanism. In inner-sphere electron transfer, the metal cofactor is oxidized or reduced by a ligand bound directly to the metal, whereas in the outer-sphere mechanism, the metal is oxidized or reduced by a group not directly coordinated to the metal cofactor. The catalysis of superoxide anions by Mn-SOD occurs by an inner-sphere mechanism. Peroxynitrite radical can react with Mn-SOD that can cause a site-specific nitration of the critical tyrosine leading to its inactivation. Four isoforms of Mn-SOD have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
