General description

The Titan One Tube RT-PCR System uses Reverse Transcriptase (RT) Avian Myeloblastosis Virus (AMV) for first strand cDNA synthesis and the Expand High Fidelity enzyme blend consisting of Taq DNA Polymerase and a polymerase with a proofreading activity for amplification of cDNA by polymerase chain reaction (PCR). The proofreading activity, leads to an increased PCR fidelity. The Titan One Tube RT-PCR System:
  • Reduces the error rate in PCR due to the proofreading capability. A threefold higher fidelity is obtained in comparision to Taq DNA polymerase.
  • Delivers increased sensitivity due to the high efficiency of all three enzymes.
  • Is faster than conventional two-step RT-PCR.

Specificity

Specificity of the reaction strongly depends on primer design and DNA contamination.
To exclude artefacts from DNA targets appropriate positive and negative control reaction should be included.

Application

Titan One Tube RT-PCR System has been used:
  • to amplify the regions containing the miRNA target (miRT) inserts/response elements in viral RNA
  • for determining the presence or absence of RNA templates
  • in cloning of rare messages without the need to construct cDNA libraries
  • for quantifying the levels of gene expression
