General description

Specificity

Clone 1F5 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Bruton tyrosine kinase (BTK). It targets an epitope within 24 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the internal region of human Bruton tyrosine kinase (BTK).

Application

Quality Control Testing

Evaluated by Western Blotting with His-tagged, full-length, recombinant, Human BTK.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected His-tagged, full-length, recombinant, Human BTK.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected BTK in lysates from Ramos cells and Rat spleen tissue.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected BTK in Ramos cells.

Affinity Binding Assay: A representative lot of this antibody bound BTK peptide with a KD of 2.9 x 10-7 in an affinity binding assay.

Enzyme Immunoassay (ELISA) Analysis: Serial dilutions from a representative lot detected His-tagged, full-length, recombinant, Human BTK in ELISA.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Tyrosine-protein kinase BTK (UniProt: Q06187; also known as EC:2.7.10.2, Agammaglobulinemia tyrosine kinase (ATK), B-cell progenitor kinase (BPK), Bruton tyrosine kinase, BTK) is encoded by the BTK (also known as AGMX1, ATK, BPK) gene (Gene ID: 695) in human. BTK is a non-receptor tyrosine kinase that is predominantly expressed in B-lymphocytes where it is involved in B cell development, differentiation, and signaling. In steady state, it is predominantly localized in the cytoplasm and following B-cell receptor (BCR) engagement by antigen, it translocates to the plasma membrane via its PH domain (aa 3-133). Its plasma membrane localization is reported to be essential for its activation. It can also shuttle between the nucleus and the cytoplasm, and its nuclear export is mediated by CRM1. BTK acts as a platform to bring together a diverse array of signaling proteins and is implicated in cytokine receptor signaling pathways. It also plays an important role in the function of immune cells of innate as well as adaptive immunity, as a component of the Toll-like receptors (TLR) pathway that links TLR8 and TLR9 to NF- B signaling pathway. BTK is activated following its phosphorylation. In primary B cells it is almost always detected in non-phosphorylated, catalytically inactive state. However, following BCR engagement and translocation to the plasma membrane, where it gets phosphorylated at tyrosine 551 by LYN and SYK, which is then followed by autophosphorylation of tyrosine 223 residue that may create a docking site for a SH2 containing proteins. Phosphorylation at serine 180 by PKC-β leads in its translocation back to cytoplasm. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
