Specificity

Clone 1O14 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Junctophilin-2 (JPH2). It targets and epitope within 23 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the C-terminal region of human Junctophilin-2 (JPH2).

Application

Quality Control Testing

Evaluated by Western Blotting in L6 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected JPH2 in L6 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected JPH2 in rat heart and human skeletal muscle tissue lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected JPH2 in human skeletal muscle and human heart tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound JPH2 with a KD of 7.0 x 10-10 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected JPH2 in L6 Cells.

Anti-JPH2, clone 1O14 ZooMAb®, Cat. No. ZRB1462, is a recombinant Rabbit monoclonal antibody that targets Junctophilin-2 (JPH2) and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Junctophilin-2 (UniProt: Q9BR39; also known as JP-2, Junctophilin type 2) is encoded by the JPH2 (also known as JP2) gene (Gene ID: 57158) in human. JPH-2 is a single-pass type IV membrane protein that is expressed in skeletal muscle and heart. It is a membrane-binding protein that provides a structural bridge between the plasma membrane and the sarcoplasmic reticulum and is required for normal excitation-contraction coupling in cardiomyocytes. JPH2 is shown to be essential for proper intracellular Ca2+ signaling in cardiac myocytes via its involvement in ryanodine receptor-mediated calcium ion release. In cardiac cells, it predominantly associates along Z lines within myocytes, whereas in skeletal muscle, it is specifically localized at the junction of A and I bands. Its transmembrane domain of JPH2 is localized in amino acids 675-695 and is anchored in sarcoplasmic reticulum membrane, while the N-terminal part associates with the plasma membrane. JPH2 can be proteolytically cleaved by calpain in response to cardiac stress and this leads to the release of the N-terminal fragment (aa 1-572). The major cleavage site is located in its C-terminal region. This N-terminal fragment serves as a transcription repressor and is required to safeguard against the deleterious effects of cardiac stress. Following cleavage and release from the membrane, it translocates to the nucleus where it binds DNA and represses expression of genes implicated in cell growth and differentiation, hypertrophy, inflammation, and fibrosis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
