General description

Transmembrane protein, 21KD (TMP21) gene is mapped on the human chromosome at 14q24.3. TMP21 belongs to the p24 family and is a type I transmembrane protein. It is located in the endoplasmic reticulum (ER) and the Golgi apparatus.

Specificity

Anti-TMP21 (N-terminal) specifically recognizes human TMP21.

Application

Anti-TMP21 (N-terminal) antibody produced in rabbit may be used in immunoblotting.

Biochem/physiol Actions

Transmembrane protein, 21KD (TMP21) has been recently linked to the metabolism of Alzheimer′s disease amyloid β (Ab) peptide. It is a member of the heteromeric secretase complex that distinctively regulates γ-secretase cleavage without affecting its ε-secretase cleavage. TMP21 is destabilized from the complex in the absence of the presenilins PS1, PS2, and pen-2. It might modulate intramembrane proteolysis controlling g-secretase activity and thus preventing the over-production of Ab peptide. Overexpression of TMP21 gene does not alter γ -secretase activity, but suppression of TMP21 gene increases Aβ40 and Aβ42 peptides. TMP21 appears to be independent of its role in protein transport from the endoplasmic reticulum (ER) to the Golgi and are involved in vesicular trafficking.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
