Serotonin transporter (SERT) and neuronal nitric oxide synthase (nNOS) interaction blocker with antidepressant efficacy in vivo.
ZZL-7 is a dipeptide that disrupts the interaction between serotonin transporter (SERT) and neuronal nitric oxide synthase (nNOS) by targeting the groove between the αB helix and βB folding of the nNOS PDZ domain. ZZL-7 decreases 293T cellular SERT-nNOS complex level (post 2h incubation at 1 μM) and demonstrates antidepressant efficacy in wt, but not nNos-/- mice in vivo (10, 20, and 40 mg/kg via intragastric administration or 10 mg/kg via i.p.) without undesirable side effects to the animals.
