General description

GCNT3 (glucosaminyl (N-acetyl) transferase 3, mucin type) is the mucin type member of core 2 β-1,6-N-acetylglucosaminyltransferase (C2GnT) family, which has three members. This gene is located on human chromosome 15q21.3, spans ~8.26kb, and has two introns and three exons. It is predominantly expressed in mucous-secreting tissues, such as small and large intestines, trachea, and stomach as well as thyroid. It is also expressed in pancreas, kidney and colon. It is a type II transmembrane protein.

Application

Anti-GCNT3 antibody produced in rabbit, a Prestige Antibody, is developed and validated by the Human Protein Atlas (HPA) project . Biochem/physiol Actions

GCNT3 (glucosaminyl (N-acetyl) transferase 3, mucin type) is responsible for the production of the three β6GlcNAc branches on secreted mucins. Not only does it catalyze the synthesis of core 2 branches, it also catalyzes the synthesis of core 4 and 1 branches. Its expression is suppressed by epidermal growth factor, and enhanced by all-trans retinoic acid and T helper 2 (Th2) cytokines such as IL-13. These cis-regulatory elements, therefore, have potential as therapeutic targets for disorders with mucus hyper-secretion. GCNT3 is responsible for the modification of O-glycans and integrin signaling, which in turn affect cell adhesion and migration and tumorigenesis. This gene is under-expressed in majority of colorectal cancer patients. It has potential to determine the relapse risk in early stage colon cancer patients, and might also help to determine the response to chemotherapy in the same.
