General description

Pollen allergen Amb t 5 (UniProt: P10414; also known as Allergen Amb t V, Allergen Ra5G, Amb t 5) binds to IgE and causes allergic reaction in human. Inntag IT10 is a small protein domain of Ambrosia trifida (Giant ragweed) that displays strong structural determinants and does not generate solubility issues in vivo and in vitro. Also, it does not compromise cell fitness when overexpressed, and has no detectable functional and localization effects when fused to a variety of target proteins. Structure of the Inntag IT10 is based on a packed beta-sheet conformation.

Specificity

Clone IT10-1 detects pollen allergen Abt t 5 in HEK293 cells transfected with GFP-IT10.

Immunogen

His-tagged recombinant peptide corresponding to 40 amino acids from Ambrosia trifida.

Anti-Inntag 10 (IT10), clone IT10 is a mouse monoclonal antibody that detects the Innocuous Inntag IT10 based on pollen allergen Abt t5 sequence and is tested for use in Immunocytochemistry and Western Blotting.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Inntag 10 (IT10) in 10 µg of transfected HEK293/GFP-IT10 cell lysates.

Target description

~33.7 kDa observed; 8.06 kDa calculated. 33.7 kD observed corresponds to 29.4 kDa GFP plus 4.3 kDa IT10. Uncharacterized bands may be observed in some lysate(s).
