Specificity

Clone 1L5 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects MYH6. It targets an epitope within 17 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal half of human MYH6.

Application

Quality Control Testing

Evaluated by Western Blotting in human heart tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MYH6 in human heart tissue lysate.

Tested Applications

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected MYH6 in human heart tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MYH6 in human heart tissue sections and human skeletal muscle tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound MYH6 peptide with at least one hundred thousand-fold (100,000X) higher than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Myosin-6 (UniProt: P13533; also known as Myosin heavy chain 6, Myosin heavy chain, cardiac muscle alpha isoform (MyHC-alpha)) is encoded by the MYH6 (also known as MYHCA) gene (Gene ID:4624) in human. MYH6 (myosin, heavy chain 6) gene encodes the heavy chain subunit of the cardiac muscle myosin, which is a hexamer made up of two heavy chain subunits, two light chain subunits, and two regulatory subunits. It is involved in the contraction of muscles. MYH6 is predominantly expressed in the heart, particularly in the atrial muscle. It is one of the major isoforms of cardiac myosin heavy chain, with its expression being more significant in the atria compared to the ventricles. MYH6 is localized in the sarcomere, the fundamental unit of muscle contraction, within the cardiac muscle cells. Specifically, it is a component of the thick filaments in the A-band of the sarcomere, where it interacts with actin filaments to facilitate muscle contraction. Defects in this gene cause atrial septal defect 3 (ASD3) characterized by incomplete closure of the wall between the atria resulting in blood flow from the left to the right atria. Its expression is also altered during thyroid hormone depletion and hemodynamic overloading. Mutations in MYH6 can disrupt the normal function of the protein, impairing cardiac muscle contraction and leading to several cardiac diseases. Mutations in this gene are associated with familial hypertrophic cardiomyopathy, dilated cardiomyopathy, and heart rhythm disorders due to malfunction of the sinus node, leading to irregular heartbeats. Genetic screening and molecular studies are crucial for identifying these mutations and understanding their impact on cardiac function. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
