General description

The TCR-CD3 multimeric complex is composed of TCRαβ, CD3γε and CD3δε and TCR‐ζ homodimer. The CD3γ, CD3δ and CD3ε, each contain one immunoreceptor tyrosine-based activation motif (ITAM), whereas the ζ chain contains three ITAMs.

Immunogen

synthetic phosphopeptide corresponding to residues surrounding Tyr142 of mouse CD3ζ, conjugated to KLH. The corresponding sequence is identical in human CD3ζ.

Application

Anti-phospho-CD3ζ (pTyr142) antibody produced in rabbit has been used in several immunochemical techniques including immunoblotting and immunoprecipitation.

Biochem/physiol Actions

Amongst the TCR subunits, the CD3 ζ chain plays a key role in regulating TCR assembly, transport, and cell surface expression. Down-regulation of TCR ζ chain expression associated with T cell dysfunction was described in various pathologies.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Adoptive cell transfer utilizing tumour-targeting cytotoxic T lymphocytes (CTLs) is one of the most effective immunotherapies against haematological malignancies, but significant clinical success has not yet been achieved in solid tumours due in part to the strong immunosuppressive tumour microenvironment.
