Biochem/physiol Actions

Cleaves the substrates consisting of consensus sequence DEVD.
Member of the CED-3 subfamily, the effector caspases, with an essential role in the apoptotic process. Upon activation, the zymogen is translocated to the mitochondrial and microsomal fractions. It is responsible for the cleavage of cellular substrates that precipitate the morphological changes of apoptosis.

Useful in screening caspase inhibitors, studying enzyme regulation, determining specificities of caspase substrates, as well as using as a positive control in caspase activity assays and Western blot analyses.

Caspases plays a key role in the execution phase of apoptosis. "Initiator" caspases, such as caspase-8, activate "effector" caspases, such as caspase-3 and -7, which subsequently cleave cellular substrates thereby precipitating the dramatic morphological changes of apoptosis.
