Target description

P-selectin (UniProt: P16109; also known as CD62 antigen-like family member P, Granule membrane protein 140, GMP-140, Leukocyte-endothelial cell adhesion molecule 3, LECAM3, Platelet activation dependent granule-external membrane protein, PADGEM, CD62P) is encoded by the SELP (also known as GMRP, GRMP) gene (Gene ID: 6403) in human. CD62P is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-41), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 42-771), a transmembrane domain (aa 772-795), and a cytoplasmic domain (aa 796-830). It serves as a calcium-dependent receptor for myeloid cells that binds to carbohydrates on neutrophils and monocytes. The ligand recognized by CD62P is reported to be sialyl-Lewis X. CD62P mediates the interaction of activated endothelial cells or platelets with leukocytes and facilitates rapid rolling of leukocyte rolling over vascular surfaces during the initial steps in inflammation through interaction with selectin P ligand (CD162). Within a few minutes following stimulation with thrombin, histamine, or phorbol 12-myristate 13-acetate (PMA), CD62P is translocated to the cell surface. (Ref.: Somers, WS., et al. (2000). Cell. 103(3); 467-479; Jutila, MA., et al. (1994). J. Immunol. 153(9); 3917-3928).

Physical form

Purified mouse monoclonal antibody IgG1 in PBS without preservatives.

Reconstitution

0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.
