General description

Cardiolipin is a unique phospholipid located in the inner mitochondrial membrane, which makes up to 20% of total lipids. It is a non-bilayer anionic phospholipid, which has four acyl chains and small headgroup.
Cardiolipins are anionic teraacylphospholipid, present in inner mitochondrial membrane eukaryotic cells and bacterial cytoplasmic membranes. 16:0 cardiolipin is synthesized by de novo pathway. 16:0 Cardiolipin binds to myeloid differentiation protein 2 (MD2).
The 16:0 number indicates the fatty acyl substituents attached to the carbons of glycerol.

Application

16:0 Cardiolipin might be suitable to assay mitochondrial fractions for CL-specific phospholipase activity.

Biochem/physiol Actions

Cardiolipin (CL) plays a key role in coordinating energy metabolism in eukaryotes. Metabolic defect in the CL pathway is implicated in dilated cardiomyopathy. CL is essential for mitochondrial functions and apoptosis. Alternations in CL content and their acyl chain composition, contributes to mitochondrial dysfunction in various pathological conditions.
Cardiolipin maintains the mitochondrial function, membrane potential and structural support for the inner mitochondrial membrane and proteins in it. Concentration and composition changes of cardiolipin have been implicated in pathological conditions including ischemia, hypothyroidism, aging, heart failure and cardioskeletal myopathy. The association of cardiolipin and cytochrome c determines the action of proteins involved in oxidative phosphorylation and apoptosis.
Cardiolipin regulates the dynamic organization of membranes and cell division. It is involved in proton uptake pathway. Cardiolipin plays an important role in mitochondrial biogenesis, fusion and fission, respiration and protein import. It modulates cell wall biogenesis, vacuole homeostasis, aging, cell cycle and apoptosis.
