Specificity

Clone 1J19 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects TLR2. It targets an epitope within 19 amino acids from the extracellular domain within the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the extracellular domain within the N-terminal half of human TLR2.

Application

Quality Control Testing

Evaluated by Western Blotting in THP-1 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TLR2 in THP-1 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected TLR2 in Human spleen tissue lysates.

Affinity Binding Assay: A representative lot of this antibody bound TLR2 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TLR2 in THP-1 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TLR2 in Human thymus tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Toll-like receptor 2 (UniProt: O60603; also known as Toll/interleukin-1 receptor-like protein 4, CD282) is encoded by the TLR2 (also known as TIL4) gene (Gene ID: 7097) in human. TLR2 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-20), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 21-588), a transmembrane domain (aa 589-609), and a cytoplasmic domain (aa 610-784). It has 19 leucine-rich repeats (LRR). It is highly expressed in peripheral blood leukocytes, bone marrow, lymph nodes, and spleen. It is also detected in lung and in fetal liver. It exists as a heterodimer with either TLR1 or TLR6 before stimulation by the ligand, and this heterodimer forms bigger oligomers in response to their corresponding ligands. It cooperates with LY96, TLR1, and TLR6 to mediate the innate immune response to bacterial lipoproteins and other microbial cell wall components. It may activate immune cells and promote apoptosis in response to the lipid moiety of lipoproteins. It acts via MYD88 and TRAF6, leading to NF-kB activation, cytokine secretion, and the inflammatory response. TLR2 is reported to serve as a receptor for M. tuberculosis lipoproteins, which ACT® as agonists to modulate antigen presenting cell function in response to the pathogen. In vitro stimulation of monocytes with phosphate-binding protein PstS1 of M. tuberculosis is reported to induce p38 MAPK and ERK1/2 activation primarily via TLR2. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
