General description
Interleukin 10 is located on human chromosome 1q32.1.
Interleukin-10 (IL-10) is a pleotropic cytokine with anti-inflammatory and immunosuppressive functions. It is the key cytokine that regulates the host inflammatory responses to bacterial, fungal and viral pathogens. The production of IL-10 is mediated by signalling of IL-6, IL-12 and TGF-β. IL-10 is secreted by a variety of cells including T cells, B cells, dendritic cells, eosinophils, natural killer cells and mast cells. The effects of IL-10 are mediated by binding to the IL-10R, consisting of α and β subunits. The signalling through IL-10R activates Jak1, Tyk2 protein kinases and JAK/STAT3 pathways. IL-10 is involved in suppression of innate immunity to kill pathogens and inhibits the release of reactive oxygen and nitrogen species from activated macrophages. Regulation of IL-10 production is important in infections, inflammatory responses in autoimmune encephalomyelitis, endotoxic shock and chronic enterocolitis
Anti-interleukin-10 antibody is specific for human IL-10. The antibody shows ~7% cross reactivity with recombinant mouse IL-10 in ELISA and immunoblotting.

Specificity
The antibody neutralizes the biological activity of recombinant human IL-10, but not recombinant mouse IL-10.

Application
Anti-interleukin 10 antibody produced in goat has been used to measure the anti-allodynic effect of IL-10 in the neuropathic pain rat model.
Anti-interleukin-10 antibody may be used for immunoblotting at a working concentration of 1 μg/ml. For ELISA a working concentration of 0.25 μg/ml is recommended. The antibody is suitable for neutralization reactions. This antibody has also been used for immunohistochemistry of cervicovaginal epithelial cells of rabbit

Biochem/physiol Actions
Interleukin 10 (IL10) helps to induce anti-tumor immune response. It controls tumor tolerance in non-small cell lung cancer. IL10 prevents the synthesis of pro-inflammatory cytokines by activating the JAK/STAT (janus kinase/signal transducers and activators of transcription) signalling pathway.
Physical form
Lyophilized from a 0.2 μm filtered solution in phosphate buffered saline containing carbohydrates.
