Specificity

Apoptosis is related to many diseases and is induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. The mammalian homologues of the key cell death gene CED-4 in C. elegans has been identified recently from human and mouse and was designated Apaf1 for apoptosis protease-activating factor 1(Zou et al. 1997; Cecconi et al. 1998). Apaf1 binds to cytochrome C (Apaf2) and caspase-9 (Apaf3), which leads to caspase-9 activation. Activated caspase-9 in turn cleaves and activates caspase-3 which is one of the key proteases, being responsible for the proteolytic cleavage of many key proteins in apoptosis (Li et al. 1997). Apaf1 can also associate with caspase-4 and caspase-8 (Hu et al. 1998). Apaf1 is ubiquitously expressed in human tissues (Zou et al. 1997).

Immunogen

Epitope: N-terminus
Polyclonal antibody raised against a peptide corresponding to amino acids 12 to 28 of human Apaf1 (Zou et al. 1997) The sequences of the immunogenic peptide are identical between human and mouse (Zou et al. 1997; Cecconi et al. 1998).

Application

Anti-Apaf-1 Antibody, N-terminus is an antibody against Apaf-1 for use in WB.

Multidomain pro-apoptotic BAX and BAK, once activated, permeabilize mitochondria to trigger apoptosis, whereas anti-apoptotic BCL-2 members preserve mitochondrial integrity. The BH3-only molecules (BH3s) promote apoptosis by either activating BAX-BAK or inactivating anti-apoptotic members. Here, we present biochemical and genetic evidence
