Specificity

Clone 1B22 is a ZooMAb^® Rabbit recombinant monoclonal antibody that specifically detects Integrin-linked protein kinase (ILK). It targets an epitope within 25 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal region of human Integrin-linked protein kinase (ILK).

Application

Quality Control Testing

Evaluated by Western Blotting in C2C12 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Integrin-linked protein kinase (ILK) in C2C12 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected ILK in HeLa, Neuro2A, and NIH3T3 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Integrin-linked protein kinase (ILK) in human kidney tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Integrin-linked protein kinase (ILK) peptide with a KD of 1.4 x 10-6 in an affinity binding assay.

Target description

Integrin-linked protein kinase (UniProt: Q13418; also known as EC:2.7.11.1, 59 kDa serine/threonine-protein kinase, Beta-integrin-linked kinase, ILK-1, ILK-2, p59ILK) is encoded by the ILK (also known as ILK1, ILK2) gene (Gene ID: 3611) in human. ILK is a member of the serine/threonine protein kinase family that is localized on the cytoplasmic side of plasma membrane and is also detected on peripheral membranes. It is highly expressed in heart followed by skeletal muscle, pancreas, and kidney. It is a receptor proximal protein kinase that regulates integrin-mediated signal transduction and serves as a mediator of inside-out integrin signaling. It undergoes rapid, but transient stimulation by cell fibronectin interactions and by insulin in a PI3-K-dependent manner, likely via the binding of PtdIns(3,4,5)P3 with a PH-like domain of ILK (aa 180-212). Its protein kinase domain is localized in amino acids 193-446 and has five ankyrin repeats within its N-terminal half. ILK is reported to phosphorylate β1 and β3 integrin subunit on serine and threonine residues. Three isoforms of ILK have been described that are produced by alternative splicing. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
