Specificity

Clone 2F8 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Talin-1. It targets an epitope within 17 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal half of human Talin-1.

Application

Quality Control Testing

Evaluated by Western Blotting in K562 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Talin-1 in K562 cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Talin-1 in Cos-7 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Talin-1 in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Talin-1 in HeLa, A431, HepG2, NIH 3T3 and Jurkat cells.

Anti-Talin-1, clone 2F8 ZooMAb®, Cat. No. ZRB1361, is a recombinant Rabbit monoclonal antibody that specifically detects Talin-1 and is tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Talin-1 (UniProt: Q9Y490) is encoded by the TLN1 (also known as KIAA1027, TLN) gene (Gene ID: 7094) in human. Talin-1 is a peripheral membrane protein that localized in adhesion complexes between cells and the extracellular matrix. It is shown to colocalize with layilin at the membrane ruffles and localized preferentially in focal adhesions than fibrillar adhesions. Talin-1 is involved in connections of major cytoskeletal structures to the plasma membrane. It binds to and activates integrins, directly connecting integrins to the actin cytoskeleton and recruiting numerous proteins, such as vinculin and focal adhesion kinase into integrin adhesion complexes. Talin-1 contains a globular N-terminal head region and a flexible rod domain, which can be dissociated by the action of calpain 2. The head region contains a FERM (protein 4.1, ezrin, radixin, moesin) domain that has binding sites for the cytoplasmic domains of β-integrins, layilin, and F-actin. The head region is also known to bind PIPK1 90 and the focal adhesion kinase. The rod domain is known to have an additional integrin-binding site, two actin-binding sites, and several binding sites for vinculin. Talin-1 expression is upregulated in several human cancers and accelerates tumor cell migration and metastasis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
