General description

Human Jak2 (GenBank Accession No. NM_004972), (amino acids. 808-end) with N-terminal His tag, MW = 42.5 kDa, expressed in Baculovirus expression system.

Application

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.

Unit Definition

One unit is defined as the amount of enzyme that will phosphorylate 1 pmol of Tyr substrate per minute at pH 7.4 and 30°C.

Physical form

Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 10% glycerol and 1 mM DTT.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.

Storage Class

12 - Non Combustible Liquids

Dysregulation of the Ras oncogene in development causes developmental disorders, "Rasopathies," whereas mutational activation or amplification of Ras in differentiated tissues causes cancer. Rabex-5 (also called RabGEF1) inhibits Ras by promoting Ras mono- and di-ubiquitination. We report here that Rabex-5-mediated
