General description

Monoclonal Anti-Kindlin-1 (mouse IgG2a isotype) is derived from the hybridoma 4A5.14 produced by the fusion of mouse myeloma cells and splenocytes from immunized BALB/c mice. Kindlin-1 is a focal adhesion protein, which belongs to the family of Kindlin proteins. It is highly expressed in epithelial cells, including keratinocytes. Kindlin-1 is localized to the focal adhesions. A splice variant potentially producing a truncated protein has been detected in the kidney, colon and small intestine. Kindlin-1 comprises a bipartite 4.1 protein, ezrin, radixin, moesin (FERM) domain with four subdomains (F0, F1, F2, and F3), which is found in many proteins. The KIND1 gene is present at human chromosome location 20p12.3.

Monoclonal Anti-Kindlin-1 recognizes human Kindlin-1.

Biochem/physiol Actions

Kindlin-1 is implicated in the activation of integrin adhesion receptors and the reorganization of the actin cytoskeleton. Deficiency of kindlin-1 is linked to Kindler syndrome, an autosomal recessive genodermatosis characterized by skin blistering, progressive skin atrophy, photosensitivity and carcinogenesis. This gene is also implicated in breast cancer, lung metastasis and lung tumorigenesis. The 4.1 protein, ezrin, radixin, moesin (FERM) domain of Kindlin-1 is involved in cytoskeletal organization. In colon cancer, Kindlin-1 is identified as a potential marker. Kindlin-1 modulates epidermal growth factor receptor signaling.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For extended storage, freeze at -20 °C in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
