General description

El glicerol, también conocido como glicerina, es un alcohol de azúcar de 3 carbonos que constituye la cadena principal de los ácidos grasos. También es un componente de los fosfolípidos y los triglicéridos. El glicerol es un líquido higroscópico de sabor dulce. El glicerol se utiliza ampliamente en formulaciones de alimentos sólidos y líquidos y de productos farmacéuticos[1] y es el principal subproducto de desecho de la producción de biodiésel a través de la transesterificación.

Application

El kit de análisis del glicerol se ha utilizado para cuantificar la liberación de glicerol de las muestras de sobrenadante del cultivo celular 3T3-L1 de ratón[2] y para medir las concentraciones plasmáticas de glicerol.[3]

Este kit es adecuado para la detección de glicerol en formulaciones de alimentos sólidos y líquidos y de productos farmacéuticos, y de muestras biológicas.

Principle

En este kit, la concentración de glicerol se determina mediante un análisis enzimático acoplado con intervención de la glicerol cinasa y la glicerol fosfato oxidasa, que da lugar a un producto colorimétrico (570 nm) o fluorométrico (ex = 535/em = 587 nm), proporcional a la glicerina presente. El rango lineal de detección de este kit es 10-1.000 μM (colorimétrico) y 2-50 μM (fluorométrico).

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. Is there any information available on how to prepare tissue for assay, considering that technical bulletin MAK117 does not include any recommendations on this matter?

    1. The general recommendation for tissue sample preparation is as follows:
      1. Begin with 20-100 mg tissue and add 200-1000 uL ice-cold PBS.
      2. Lysis can be achieved by homogenization (10-20 passes in a Dounce homogenizer on ice) or by sonication (preferably performed in an ice-water bath). The degree of tissue lysis can be checked under a microscope.
      3. Centrifuge the homogenate at 14,000 g for 10 min.
      4. Transfer the clear supernatant into a clean tube and use it for the assay.

  3. Can serum glycerol content be analyzed or just plasma?

    1. Yes, this product can determine glycerol content in serum samples.

  4. How long are reagents stable at 2-8 C after initial thaw?

    1. The Assay Buffer, MAK117A, may be stored at 2–8 Celsius. For all other components, it is recommended to aliquot and freeze at - 20 deg C. Avoid repleated freeze/thaw cycles.
