Descripción general

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application. This ColorWheel® Dye-Ready monoclonal antibody was developed using our proprietary ZooMAb® recombinant antibody technology. It offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Especificidad

Clone 1C22 is a rabbit monoclonal antibody that detects Histone H2AX. It targets an epitope within 15 amino acids from the C-terminal region.

Tested Applications

Histone H2AX (UniProt: P16104; also known as H2a/x, Histone H2A.X) is encoded by the H2AX (also known as H2AFX) gene (gene ID: 3014) in human. Histones are highly conserved basic nuclear proteins that are responsible for the nucleosome structure of chromatin in eukaryotes. They play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which DNA is wrapped in repeating units, called nucleosomes, which limits DNA accessibility to the cellular machineries that require DNA as a template. The histone H2A.X is a variant member of the H2A family of histones that is distinguished from other H2A histones by a unique carboxy-terminal sequence. This unique sequence is highly conserved throughout eukaryotic evolution and is rapidly phosphorylated by ATM or ATR at serine 139 in mammals in response to DNA double-strand breaks. H2A.X phosphorylation is important in the formation of a stable repair complex at the site of DNA damage. Phosphorylation of H2A.X is important in the formation of a stable repair complex at the site of DNA damage. This phosphorylation is a very rapid response to DNA damage, occurring within as little as one minute after exposure to ionizing radiation. (Ref.: Paull, TT., et al. (2000). Curr. This ColorWheel® Dye-Ready monoclonal antibody was developed using our ZooMAb® recombinant monoclonal antibody (Cat. No. ZRB1663). It offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Forma física

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitución

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Almacenamiento y estabilidad

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
