General description

Src-like-adapter 2 (UniProt: Q9H6Q3; also known as Modulator of antigen receptor signaling; MARS; Src-like adapter protein 2; SLAP-2) is encoded by the SLA2 (also known as SLAP2, C20orf156) gene (Gene ID 84174) in human. SLAP-2 is localized to the plasma membrane and intracellular vesicles, including late endosomal vesicles. The SLAP-2 protein contains an NH2-terminal myristoylation consensus sequence and SH3 and SH2 Src homology domains, but lacks a tyrosine kinase domain. It is predominantly expressed in immune system, including T cells and monocytes and at low levels in B cells, placenta, prostate, colon, and skin. SLAP-2 serves as an adapter protein that negatively regulates T-cell receptor (TCR) signaling and is shown to block T-cell antigen-receptor induced activation of nuclear factor of activated T-cells. In antigen receptor-stimulated cells, SLAP-2 associates with several tyrosine phosphorylated proteins, including the ubiquitin ligase Cbl. Deletion of the carboxyl terminus of SLAP-2 is shown to block its function and abolish its association with Cbl. Overexpression of SLAP-2 in T and B cell lines impairs antigen receptor-mediated signaling events, including the up-regulation of CD69, NFAT promoter activation, and calcium influx. Clone Z30P1F12*F4 detects -Src-Like-Adaptor 2 (SLA2) protein in human by targeting a sequence in the C-terminal region.
