General description

The protein named Anoctamin-2 or alternatively Transmembrane protein 16B and encoded by the human gene ANO2/C12orf3/TMEM16B is a calcium activated chloride channel that plays a role in smell or olfactory signal transduction and photoreception. Odorant molecules bind to odor-sensing receptors leading to an increase in calcium entry that then activates TMEM16B calcium current which amplifies the depolarization of the Odor-sensing receptors. TEMEM16B also mediates light perception amplification in the retina where TMEM16B is a component of the presynaptic protein complex recruited to specialized plasma membrane domains of photoreceptors. TEMEM16B is expressed in sensory neurons particularly in the retina and olfactory neurons; it localizes to the plasma membrane where it is a multi-pass membrane bound protein. TEMEM16B has multiple isoforms and recent research suggest that the native channel may be composed of two or more splicing isoforms acting as subunits that together shape the channel properties.
