General description

N-methyl-D-aspartate receptor subunit NR1 (NMDAR1) is a multi-pass membrane protein belonging to the glutamate-gated ion channel family and is one of two subunits comprising the tetrameric protein NMDAR localized throughout the brain at excitatory synapses. NMDAR1 is mediated by glycine and is a critical component to excitotoxicity, learning, memory acquisition, synaptic plasticity, and synaptogenesis. It also plays a regulatory role for neuronal functions involving glutamate neurotransmission and the targeting of NMDA receptors on the cell surface. As a glutamate-gated ion channel, NMDAR1 is highly calcium permeable and possesses sensitivity to magnesium in a voltage-dependent manner.

Immunogen

Western Blot Analysis: 0.1 µg/mL of this antibody from a previous lot detected NMDAR1 on 10 µg of mouse microsomal brain tissue lysate.

Immunohistochemistry Analysis: 1:250 dilution of this antibody from a previous lot detected NMDAR1 in rat cerebellum tissue.

Quality

Evaluated by Western Blot in rat brain tissue lysate.

Western Blot Analysis: 0.1 µg/mL of this antibody detected NMDAR1 on 10 µg of rat brain tissue lysate.

Target description

~120 kDa observed

Physical form

Protein A
Purified rabbit monoclonal IgG in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide and 40% Glycerol.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.

Analysis Note

Control
Rat brain tissue lysate

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.
