General description

The Ca2+/Calmodulin-Dependent Kinase family includes CaM Kinases I through IV, Phosphorylase Kinase, and MLCK. The CaM Kinases are inactive in the absence of stimuli; binding to Ca2+/Calmodulin induces conformational changes and activation. CaM Kinases have broad substrate specificity, with many cellular functions.

Specificity

Recognizes proteins of 50-72 kDa, corresponding to apparent molecular mass of CaMKII isoforms on SDS-PAGE immunoblots. Recombinant rat calmodulin-dependent protein kinase II α subunits expressed in sf9 insect cells are also detected. As the sequences of the rat α, δ, and γ isoforms are conserved over this amino terminus region, this antibody is expected to recognize the δ, γ, and α isoforms. Antibody is not expected to cross-react with the β isoform. Proteins of unknown identity may be detected on immunoblot analysis with some lysates (40, 45 and 90 kDa).

Immunogen

Epitope: N-terminus
Synthetic peptide corresponding to the N-terminus of Rat CaMK II

Application

Anti-CaM Kinase II Antibody is an antibody against CaM Kinase II for use in WB.
Optimal working dilutions must be determined by end user.

Quality

Routinely evaluated by Western Blot on HEK293 lysates.

Western Blot Analysis:
1:500 to 1:1,000 dilution of this lot detected CaM Kinase II on 10 μg of HEK293 lysates.

Target description

50-72 kDa, corresponding to apparent molecular mass of CaMKII isoforms.

Linkage

Replaces: AB3111

Physical form

Affinity purified
Purified rabbit polyclonal IgG in buffer containing borate buffered saline and phosphate buffered saline containing 0.05% sodium azide.

Storage and Stability

Stable for 1 year at -20°C in undiluted aliquots from date of receipt. For optimal storage, aliquot and store at -20°C or -70°C.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Control
HEK293 cell lysate
