Specificity

Clone 2E21 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects MKK3. It targets an epitope within 15 amino acids from the C-terminal half. It also displays reactivity with MKK6.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal region of human MKK3.

Application

Quality Control Testing

Evaluated by Western Blotting in HEK293 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected MKK3/MKK6 in HEK293 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected MKK3/MKK6 in Caco2, C2C12, and C6 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MKK3/MKK6 in Caco-2 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MKK3/MKK6 in Human small intestine tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Dual specificity mitogen-activated protein kinase kinase 3/6 (UniProt: P46734/ P52564; also known as EC:2.7.12.2, MAP kinase kinase 3/6, MAPKK 3, MAPK/ERK kinase 3/6, MEK 3/6, Stress-activated protein kinase kinase 2, SAPK kinase 2/3, SAPKK-2/3, SAPKK2/6) is encoded by the MAP2K3/6(also known as MEK3/6, MKK3/6, PRKMK3/6, SKK2/3) gene (Gene ID: 5606/5608) in human. MKK3/MKK6 are dual specificity kinases that act as essential components of the MAP kinase signal transduction pathways. They catalyze  the phosphorylation of a threonine and a tyrosine residue in MAP kinases, such as MAPK14/p38-MAPK.  Both MKK3 and MKK6 are known to phosphorylate p38 MAP kinase but do not phosphorylate the related JNK or ERK MAP kinases. Their protein kinase domain is localized in amino acids 64-325 (MKK3) and 53-314 (MKK6). Clone 2E21 is generated using a sequence within the protein kinase domain that is common to both MKK3 and MKK6. MKK3 phosphorylation on serine 218 and threonine 222 by MAP kinase kinase kinases positively its kinase activity.  MKK6 is phosphorylated at serine 207 and threonine 211 in response to a variety of cellular stresses, including UV radiation, osmotic shock, hypoxia, and inflammatory cytokines. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
