Specificity

Clone 1O12 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects MDR1 protein. It targets an epitope within 22 amino acids from internal, cytoplasmic loop region.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the internal cytoplasmic loop region of human Multidrug resistance protein 1 (MDR-1).

Application

Anti-MDR-1, clone 1O12 ZooMAb, Cat. No. ZRB1521, is a recombinant Rabbit monoclonal antibody that targets MDR-1 and is tested in Affinity Binding Assay, Flow Cytometry, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MDR-1 in HepG2 and NIH 3T3 cells.

Affinity Binding Assay: A representative lot bound MDR-1 peptide with a KD of 3.5 x 10-7 in an affinity binding assay.

Flow Cytometry Analysis: 0.01 μg from a representative lot detected MDR-1 in HepG2 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected MDR-1 in human adrenal gland tissue sections.

Target description

Multidrug resistance protein 1 (UniProt: P08183; also known as ATP-binding cassette sub-family B member 1, ATP-dependent translocase ABCB1, P-glycoprotein 1, Phospholipid transporter ABCB1, CD243; MDR1) is encoded by the ABCB1 (also known as MDR1, PGY1) gene (Gene ID: 5243) in human. In tumor cells, the classical MDR phenotype is conferred by MDR1 that is expressed in about 50% of human cancers. MDR1 is a multi-pass membrane N-linked glycoprotein that translocates drugs and phospholipids across the membrane. It contains 6 extracellular domains, 12 transmembrane domains, and 7 cytoplasmic domains. Its ABC transporter domains are localized to amino acids 392-628 and 1035-1273.MDR1 expression has been reported in liver, kidney, small intestine, and brain. It catalyzes the flop of phospholipids from the cytoplasmic to the exoplasmic leaflet of the apical membrane. It participates mainly to the flop of phosphatidylcholine, phosphatidyl-ethanolamine, beta-D-glucosylceramides and sphingomyelins. It serves as an energy-dependent efflux pump that is responsible for decreased drug accumulation in multidrug-resistant cells. Overexpression of MDR1 is shown to be of prognostic value in leukemia, neuroblastoma and sarcoma and its inhibition can restore chemosensitivity in tumor cells. Mutations in ABCB1 gene has been linked to inflammatory bowel disease 13, a chronic, relapsing inflammation of the gastrointestinal tract with a complex etiology. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

0.3 mg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
