Gene Information

General description

WASP homologue associated with actin, membranes, and microtubules (WHAMM) is a nucleation promoting factor. It is located in the cis-Golgi and tubulo-vesicular membrane transport intermediates. WHAMM contains an N-terminal region, a coiled-coil region and a WCA region. It is located on the human chromosome at 15q25.2.

Specificity

Anti- WHAMM (C-terminal) recognizes human WHAMM.

Immunogen

peptide corresponding to the C-terminal region of human WHAMM , conjugated to KLH.

Application

Anti-WHAMM (C-terminal) produced in rabbit may be used in immunoblotting.

Biochem/physiol Actions

WASP homologue associated with actin, membranes, and microtubules (WHAMM) takes part in actin-related protein 2 & 3 (Arp2/3)-mediated actin polymerization, mediates Golgi membrane association and microtubule dynamics. It also takes part in the maintenance of Golgi structure and participates in vesicle transport between the endoplasmic reticulum and the Golgi apparatus.

Physical form

Solution in 0.01 M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2–8 °C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.

The Arp2/3 complex is an actin nucleator that plays a critical role in many cellular processes. Its activities are regulated by nucleation-promoting factors (NPFs) that function primarily during plasma membrane dynamics. Here we identify a mammalian NPF called WHAMM (WASP
