General description

ERLIN1 (ER lipid raft associated 1) is a member of the prohibitin protein family, and contains the ~160 amino acid long prohibitin-homology domain (PHB). It has a molecular weight of ~40kDa. It exists in association with lipid rafts, and is found in the ER (endoplasmic reticulum). ERLIN1 mRNA is expressed in a wide range of human tissues. This protein contains a transmembrane N-terminal, which is responsible for targeting this protein to the ER, and a C-terminal containing the SPFH (Stomatin, Prohibitin, Flotillin, and HflC/HflK) domain, projecting into the ER lumen. ERLIN1 forms a hetero-oligomeric complex with ERLIN2, with which it shares high sequence homology.

Application

Biochem/physiol Actions

ERLIN1 (ER lipid raft associated 1) is differentially expressed in activated and non-activated dendritic cells (DCs), and might play a role in the activation of DCs. ERLIN1/ERLIN2 complex is involved in the endoplasmic reticulum (ER) associated degradation (ERAD) of inositol 1,4,5-trisphosphate (IP3) receptor, post IP3-receptor activation. Variants in the ERLIN1-CHUK-CWF19L1 gene cluster are associated with simple steatosis and hepatic steatosis with inflammation. Thus, this cluster might influence the susceptibility to non-alcoholic fatty liver disease (NAFLD). ERLIN1 regulates and inhibits sterol regulatory element binding proteins (SREBPs) during cholesterol sufficiency, and therefore, contributes to cellular cholesterol homeostasis.
