General description

Ceramide synthesis is majorly driven by de novo pathway involving the palmitoyl-CoA and L-serine, and also by salvage pathway via recycling of complex sphingolipids.[1] The ceramide transporter, Cert mediates the transfer of C14 from smooth endoplasmic reticulum to Golgi.[1] Ceramide synthase CerS6 enzyme is involved in the synthesis of C14 ceramide.[2]

Application

C14 Ceramide (d18:1/14:0) is suitable for use
  • as a standard in high-performance liquid chromatography/tandem mass spectrometry (LC/MS2) for ceramide quantification from muscel biopsies[3]
  • in lipid monolayer preparation for biophysical studies[4]
  • in the morphological and dimensional evaluation studies[5]

Biochem/physiol Actions

Ceramides regulate biological processes including proliferation, differentiation and apoptosis.[5] In the cystic fibrosis patients, elevated levels of C14 ceramide is observed.[1] It is also a potential biomarker for hepatic steatosis (HS) and C14 ceramide levels correlate with the aspartate aminotransferase (AST) / alanine transaminase (ALT) ratio.[6] Tumor necrosis factor α (TNFα) mediates increase in the C14 levels. Fumonisin B1 prevents the C14 accumulation.[2]
