Specificity

Clone 5C15 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Hematopoietic progenitor cell antigen CD34. It targets an epitope within 15 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal region of human Hematopoietic progenitor cell antigen CD34.

Application

Anti-CD34, clone 5C15 ZooMAb, Cat. No. ZRB1232, is a recombinant Rabbit monoclonal antibody that specifically targets CD34 and has been tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CD34 in human kidney tissue lysaste.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CD34 in human kidney and rat kidney tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD34 in PC3 cell line.

Flow Cytometry Analysis: 1 µg of a representative lot of this antibody detected CD34 in one million fixed and permeabilized KG-1 cells.

Note: Actual optimal working dilutions must be determined by the end user as specimens and experimental conditions may vary.

Target description

Hematopoietic progenitor cell antigen CD34 (UniProt: P28906; also known as CD34) is encoded by the CD34 gene (Gene ID: 947) in human. CD34 is a highly glycosylated single-pass type I membrane protein that is selectively expressed on hematopoietic progenitor cells and the small vessel endothelium of a variety of tissues. It is predominantly regarded as a marker of hematopoietic stem cells and hematopoietic progenitor cells. L-Selectin (CD62L) is the most commonly described ligand for CD34. CD34 serves as an adhesion molecule that plays a role in early hematopoiesis by mediating the attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells. It is also reported to act as a scaffold for the attachment of lineage specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components. Under normal conditions, CD34+ expressing cells account for about 1 to 2% of the total bone marrow cells. CD34 is synthesized with a signal peptide (aa 1-31), which is subsequently cleaved off to produce the mature form. The mature form contains an extracellular domain (aa 32-290), a transmembrane domain (aa 291-311), and cytoplasmic domain (312-385). The extracellular domain that is heavily sialylated and O-linked glycosylated and contains some N-linked glycosylation sites. Two isoforms of CD34 have been described that are produced by alternative splicing. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
