General description

Zinc finger protein castor homolog 1 (UniProt Q9CWL2; also known as Castor-related protein, Putative survival-related protein, Zinc finger protein 693) is encoded by the Casz1 (also known as Cst, D4Ertd432e, Kiaa3026, Srg, Znf693) gene (Gene ID 69742) in murine species. Originally identified as a Drosophila caster gene product involved in neural fate determination, mammalian CASZ1 protein plays a critical role during the development of a variety of different cell and tissue types, including brain and heart. CASZ1 directly targets and promotes EGFL7 transcription in endothelial cells (ECs), thereby stimulates EGFL7/RhoA-mediated vessel assembly and morphogenesis. Both human and murine express two spliced CASZ1 isoforms/variants (CASZ1a/CASZ1V1 and CASZ1b/CASZ1V2; also known as hCASZ11 and hCASZ5 in human), with the larger isoform containing eight C2H2-type zinc fingers and the short isoform containing only the first four zinc fingers. Loss of CASZ1 expression is found in neuroblastoma tumors of patients with a poor prognosis, while restoration of either CASZ1a/CASZ1V1 or CASZ1b/CASZ1V2 suppresses neuroblastoma tumor growth in cultures. In addition, CASZ1b/CASZ1V2 is reported to bind histones and interact with NuRD (nucleosome-remodeling deacetylase) complex, as well as DNA repair proteins. Disruption of either H3/DNA repair protein or NuRD binding to CASZ1b/CASZ1V2 significantly decreased its transcriptional activity.

Specificity

This guinea pig polyclonal antiserum stained only CASZ1-expressing, but not Casz1 gene-knockout cells in retina tissue sections from E17.5 mouse embryons (Courtesy of Pierre Mattar, Christine Jolicoeur, and Michel Cayouette, Institut de recherches cliniques de Montreal (IRCM), Canada). Epitope regions are present in both CASZ1 spliced isoforms/variants (CASZ1V1 and CASZ1V2) of human and mouse protein reported by UniProt (Q86V15 and Q9CWL2). Human spliced isoforms/variants 1 and 2 are also known as hCASZ11 and hCASZ5, respectively.

Immunogen

A mixture of three peptides representing two internal sequences N-terminal to the first zinc finger and a third sequence within the region between the third and fourth zinc fingers.

Application

Anti-CASZ1, Cat. No. ABN1727, is a highly specific guinea pig polyclonal antibody that targets Zinc finger protein castor homolog 1 and has been tested in Immunocytochemistry, Immunofluorescence, Immunoprecipitation, and Western Blotting.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot stained 4% paraformaldehyde-fixed, 0.4% Triton X-100-permeabilized NIH/3T3 cells transfected to express either of the two mouse CASZ1 spliced variants by fluorescent immunocytochemistry (Courtesy of Pierre Mattar, Christine Jolicoeur, and Michel Cayouette, Institut de recherches cliniques de Montreal (IRCM), Canada).

Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected CASZ1 immunoreactivity only among EYFP-negative cells, but not EYFP-positive Casz1-knockout cells by fluorescent immunohistochemistry staining of 4% paraformaldehyde-fixed retina cryosections from E17.5 transgenic mouse embryos carrying floxed Casz1 alleles and a ROSA26-floxed-stop::EYFP reporter allele, as well as Pax6 promoter-driven cre recombinase expression to allow tissue-specific (conditional) Casz1 knockout (Courtesy of Pierre Mattar, Christine Jolicoeur, and Michel Cayouette, Institut de recherches cliniques de Montreal (IRCM), Canada).

Western Blotting Analysis: A 1:5,000 dilution of this antiserum detected both CASZ1 spliced variants (CASZ1V1 and CASZ1V2) of either human or mouse origin in 10 µg of lysates from respective HEK293T transfectants.

~190/125 kDa observed. 190.1/124.8 kDa (human isoform 1/2), 191.2/125.6 kDa (mouse isoform 1/2) calculated. Uncharacterized bands may be observed in some lysate(s).

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
