General description

SDF-1α (stromal cell-derived factor 1 α) and β are stromal derived CXC chemokines, and signal through the CXCR4 (C-X-C motif chemokine receptor). SDF1α is an isoform of CXCL12 (C-X-C motif chemokine 12, also referred to as SDF-1). The gene is mapped to human chromosome 10q11. Recombinant human SDF-1a is an 8.0kDa protein containing 68 amino acid residues.

Application

SDF-1α (stromal cell-derived factor 1 α) human has been used to study the role of SDF-1α in cell migration and survival after TGF (transforming growth factor)-β-induced EMT (epithelial–mesenchymal transition) in hepatoma cells. It has been used to study podia formation, migration, proliferation and cell-cell adhesion of human hematopoietic progenitor cells in presence of SDF-1α.
SDF-1α (stromal cell-derived factor 1 α) human has been used in the chemotaxis assay of neural crest cells (NCCs) and as a component of Dulbecco′s modified Eagle′s medium (DMEM) in migration assay of human mesenchymal stem cells (hMSCs).

Biochem/physiol Actions

SDF-1α (stromal cell-derived factor 1 α) and β chemoattract B and T cells, and have been shown to induce migration of CD34+ stem cells. SDF-1 is mainly expressed in the inflamed and failing myocardium. It is involved in inflammation, proliferation, tumorigenicity and metastasis. The SDF-1 proteins exert HIV (human immunodeficiency virus) suppressive activity in cells expressing the CXCR4 (C-X-C motif chemokine receptor 4). It is upregulated in breast carcinoma, ovarian carcinoma and papillary thyroid carcinoma. It enhances angiogenesis in the ischemic tissue. SDF1 is also involved in neuroprotection after ischemic stroke with the help of CXCR4.

Lyophilized with no additives.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
