General description

Dinitrophenyl (DNP) is a hapten that is often used for labeling primary or secondary probes in immunological assays. Hence, anti-DNP antibody is a useful tool for the detection and analysis of target molecules. Monoclonal Anti-DNP antibody is specific for DNP.
IgE gene is encoded on human chromosome 14. IgE is the least circulating antibody, but critical for eliciting hypersensitivity reaction against allergens.

Application

Applications in which this antibody has been used successfully, and the associated peer-reviewed papers, are given below.

Monoclonal Anti-Dinitrophenyl antibody is suitable for use in flow cytometry and immunocytochemistry.
Monoclonal antidinitrophenyl specific mouse IgE has been used in degranulation of rat mast cells using microplate reader and fluorescence spectrophotometer.
The antibody may be used for experimental induction of controlled allergic reactions or as a source of mouse IgE.
The antibody may be used for experimental induction of controlled allergic reactions or as a source of mouse IgE.To induce systemic anaphylaxis, mice were injected with 3 μg of anti-DNP mouse IgE and then given by IV 500 μg of DNP-BSA twenty hours later. To induce local anaphylaxis, mice were injected intradermally with 10ng of anti-DNP mouse IgE and then challenged with an i.v. injection of 500 μg DNP-BSA.

Biochem/physiol Actions

IgE is produced by B cells by class switch recombination in response to allergens. IgE is also associated with mast cells and is abundantly produced in response to allergens especially in asthma and allergic rhinitis. IgE regulates mast cell homeostasis and is upregulated in food allergy. IgE is implicated in the pathogenesis of autoimmune diseases like rheumatoid arthritis and systemic lupus erythematosus. IgE levels are modulated by other genes present on chromosome 14 especially in patients with asthma. Patients with mutations in signal transducer and activator of transcription 3 (STAT3) are associated high levels of IgE called the hyper–immunoglobulin E syndrome (HIES).

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, the product may be stored at 2-8 °C for up to one month. For extended storage, freeze at -20 °C in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
