General description

ULK1 or autophagy-related protein 1 homolog (ATG1) is a serine/threonine protein kinase. It is a ubiquitously expressed protein that is highly abundant in the brain, skeletal muscle, and heart. It is involved in the signaling pathway of mTOR receptors leading to the regulation of autophagy in nutrient-deprived cells. mTOR may interact with ULK1 as a complex of ULK1/FIP200/Atg13. ULK1 may play an important role in neuronal development: Elimination of ULK1′s enzymatic activity by targeted disruption of its kinase domain has blocked the outgrowth of neurons in the developing mouse brain, possibly by blocking the ULK1-mediated trafficking of neurotrophins.

Specificity

This antibody recognizes ULK1 phosphorylated at Ser555.

Epitope: Phosphorylated Ser555
KLH-conjugated linear peptide corresponding to human ULK1 phosphorylated at Ser555.

Application

Peptide Inhibition Analysis: A representative lot of this antibody blocked ULK1 in PIA involving peptide 3123-C vs. peptide 3123 on serum starved MCF7 cell lysates.

This Anti-phospho-ULK1 (Ser555) Antibody is validated for use in Western Blotting, Peptide Inhibition Assay for the detection of phospho-ULK1 (Ser555).

Quality

Evaluated by Western Blot in non-serum starved and serum starved MCF7 cell lysates.

Western Blot Analysis: 0.2 µg/mL of this antibody detected phospho-ULK1 (Ser555) in 10 µg of non-serum starved and serum starved MCF7 cell lysates.

Target description

~120 kDa observed. Uniprot describes a molecular weight at 112 kDa This protein has been observed at greater than ~125 kDa due to multiple phosphorylation-dephosphorylation processes. (Ganley, I. G., et al. (2009). J Biol Chem. 284(18):12297-12305.)
