Specificity

Target description

G-protein coupled receptor 161 (UniProt: B2RPY5; also known as GPR161) is encoded by the Gpr161 (also known as Gm208) gene (Gene ID: 240888) in murine species. GPR161 is a multi-pass membrane glycoprotein of the GPCR family with seven transmembrane domains. It localizes to the ciliary membrane and is also detected in the recycling endocytic compartment. It is ubiquitously expressed from 8.5 dpc and is mostly concentrated in the developing nervous system at later stages. By 10.5 dpc, it is mainly expressed in the neural tube. At later embryonic stages it is predominantly expressed in the brain, spinal cord, and dorsal ganglia. GPR161 functions as a negative regulator of sonic hedgehog (Shh) signaling, which promotes the processing of Gli3 into its repressive form (Gli3R) during neural tube development via cAMP-protein kinase A signaling. In the presence of Shh, it is removed from primary cilia and is internalized into recycling endosomes, preventing its activity, and allowing activation of the Shh signaling. Gpr161 knockout mice display increased Shh signaling and exhibit embryonic lethality by 10.5 dpc. GPR161 is overexpressed in triple-negative breast cancer and serves is a regulator of mammary epithelial cell proliferation and invasion in a mTOR-dependent manner. Knockdown of GPR161 is reported to impair proliferation of human basal breast cancer cell lines and its overexpression increases cell proliferation, migration, and intracellular accumulation of E-cadherin. Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
