Biochem/physiol Actions
A2M (α-2-macroglobulin) inhibits all four classes of proteinases by a unique ′trapping′ mechanism. This protein undergoes a conformational change when a proteinase binds and cleaves a specific peptide stretch called the "bait region" in the protein. This modified protein traps the bound proteinase It is involved in the clearance and degradation of Aβ, which is a major component of beta-amyloid deposits. Therefore, it is associated with Alzheimer′s disease (AD).
