General description

GAP-43, a common marker of differentiating neurons, is expressed at elevated levels by developing or regenerating neurons during axon growth. While GAP-43 is an integral membrane protein associated with the cytoplasmic surface of axonal growth cones (and synapses), it is absent from dendritic growth cones. In adult brain, GAP-43 is found in high concentration in presynaptic areas where memory formation is thought to occur, such as frontal cortex, limbic system and hippocampus. Phosphorylation of GAP-43 by PKC (at Ser41 on human, mouse, rat, and bovine GAP-43 or Ser42 on chicken and Xenopus GAP-43) is specifically correlated with certain forms of synaptic plasticity.

Specificity

Reacts to a lesser degree with rat, human, bovine and chicken. Other species have not been tested.
Recognizes Growth Associated Protein-43 (GAP-43).

Immunogen

Recombinant rat GAP-43 (complete sequence).

Application

Anti-Growth Associated Protein-43 (GAP-43) Antibody is an antibody against Growth Associated Protein-43 (GAP-43) for use in ELISA, IC, IH, IP & WB.
Immunohistochemistry:
1:1,000 dilution of a previous lot was used on 4% paraformaldehyde / PBS fixed or 3% Paraformaldehyde / 0.1% Gluteraldehyde / PBS fixed tissue sections.

Immunocytochemistry:
1:1,000 dilution of a previous lot was used on Hippocampal neurons in culture and PC12 cells stimulated with NGF.

Immunoprecipitation:
1:100 dilution of a previous lot was used.

ELISA:
1:2,000 dilution of a previous lot was used.

Quality

Routinely evaluated by Western Blot on Mouse brain lysates.

Western Blot Analysis: 1:500 dilution of this lot detected Growth Associated Protein 43 on 10 μg of Mouse brain lysates.

Purified rabbit polyclonal in buffer containing PBS with 0.01% sodium azide as a preservative.

Storage and Stability

Stable for up to 6 months at -20ºC in undiluted aliquots from date of receipt.
Avoid repeated freeze/thaw cycles. During shipment, small volumes of product will occasionally become entrapped in the seal of the product vial. For products with volumes of 200 µL or less, we recommend gently tapping the vial on a hard surface or briefly centrifuging the vial in a tabletop centrifuge to dislodge any liquid in the container’s cap.
