Specificity

Clone 3C3 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Topoisomerase II. It targets an epitope within 20 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the C-terminal region of human Topoisomerase II.

Application

Quality Control Testing

Evaluated by Western Blotting in Neuro2a cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Topoisomerase II in Neuro2a cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Topoisomerase II in HeLa cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Topoisomerase II in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Topoisomerase II in A431, HeLa, HepG2, and NIH3T3 cells.

Affinity Binding Assay: A representative lot of this antibody bound Topoisomerase II peptide with a KD of 5.7 x 10-7 in an affinity binding assay.

Target description

DNA topoisomerase 2-alpha (UniProt: P11388; also known as EC:5.6.2.2 1, DNA topoisomerase II, alpha isozyme) is encoded by the TOP2A (also known as TOP2) gene (Gene ID: 7153) in human. DNA topoisomerases are nuclear enzymes that regulate the conformational changes in DNA topology by catalyzing the breakage and rejoining of DNA strands during the normal cell cycle. They relieve torsional stress during replication and transcription. Three different types of topoisomerases have been reported in human. They are types 1; 2 , and 2β. They are encoded by genes on three separate chromosomes. Simpler organisms possess only topoisomerase 1; however, higher organisms have all three types of topoisomerases. Topoisomerase I reversibly cleaves a single strand in duplex DNA molecule, whereas topoisomerase 2 breaks and rejoins both DNA strands. Topoisomerase II is found at replication forks and remains tightly associated with chromosomes during mitosis. The N-terminus of Topoisomerase 2 contains several structural domains; the ATPase domain (about residues 1-265), the transducer domain (about 266-428) and the toprim domain (455-572). It is essential for the survival of actively growing cells. Its levels are upregulated dramatically during periods of cell proliferation. Its levels increase over the cell cycle and peak in G2/M phase. Topoisomerase II functions in growth-dependent processes, such as DNA replication and chromosome segregation. In contrast, the expression of the β isoform is independent of proliferative status and it dissociates from chromosomes during mitosis. Topoisomerase IIβ cannot compensate for the loss of topoisomerase II in mammalian cells. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
